Carbolfuchsin, also known as carbol fuchsin or carbol-fuchsin, is a staining solution commonly used in microbiology, histology, and cytology for the visualization of acid-fast bacteria, particularly Mycobacterium species such as Mycobacterium tuberculosis. It is a mixture of basic fuchsin dye and phenol or carbol, giving it a dark red to purple color. The addition of phenol or carbol to fuchsin enhances its penetration into the waxy cell walls of acid-fast bacteria, making them resistant to decolorization by acid-alcohol during staining procedures. This property allows acid-fast bacteria to retain the red stain while other non-acid-fast bacteria are decolorized, aiding in their differentiation under a microscope. Carbolfuchsin is a critical component of the Ziehl-Neelsen and Kinyoun staining methods, which are widely used for the detection of acid-fast bacteria in clinical and research settings.
